What would happen if I fire a 5.56 round trough a .223 barrel ??
it's not the barrel, it's whether or not the chamber is rated for 5.56. 5.56 ammo usually has a higher pressure, like firing a +p or +p+ round through a firearm that is not rated for +p or +p+ loads. What kind of firearm are you asking about?

I just received a Armalite upper 16” M15A4 complete upper assembly with short hand guard heavy barrel.
There are no stamps on the barrel for caliber, now I don’t know what is it .223 or 5.56 ???

Did you also check the barrel under the handguard? Sometimes, it's there.

POF lower I have shot it with .223 but not 5.56 yet

I'm pretty sure Patriot is manufactured to Mil-spec, thus 5.56 is perfectly ok. It's odd that there are no markings on th barrel, did you buy the upper new?
It's the upper that matters, not the lower. Anyway, I have 2 POF's, shooting both the 5.56 and .223.

Yes I got the upper new from Armalite
