Can 00 buck be fired effectively through rifled barrels? Will it damage the rifling?

Never having done this with a 12gauge, I truly don't know. That being said, they make shot shells for .22lr, 9mm, etc...and now the Taurus Judge that has .45lc and .410 capability.
I would say that one could safely fire 00 buckshot from their rifled barrel; however, I'd recommend cleaning the barrel more than normal to help prevent the lead build up, and I wouldn't make it a habbit of firing the buckshot from the rifled barrel as it probably would damage the rifling over time.
Depending on what the intended purpose behind the rifled barrel is, you may be better served with a smooth bore barrel with the Vang Comp treatment for shooting both buckshot and slugs.

It won't hurt the rifling, you'll need to clean it more though. I've shot OO out of rifled barrels and it didn't hurt anything other than the pattern, still suitable for close range work. I'll have to get the digital camera out sometime and pattern my rifled barrel.
