I am taking my girls on a cruise at the end of July. We will be driving from Tennessee to Lousiana to get to the port. I know I can't take my gun on the ship with me. I don't want to travel without it but I'm not sure I want to leave it in the car at the port either. I'm not sure what I should do. What do yall think?

Are you sure you can't take it onboard? Ships don't have all the security screening that planes have.
Seriously, I would NOT leave it in my vehicle parked in Lousiana.
You might ask your travel agent or the cruise ship company if they offer any kind of rental lockers for valuables you can leave on shore. But I would not mention the fact that it is a gun.

Cruise documents say "no weapons" so I know I can't take it on board. The parking is secured and patrolled so I might just have to risk it.

I agree with asking the cruise ship about a secure locker for a firearm. Offer to have them lock it up when arriving, and give it back as you exit the ship. I'd hate to be traveling between states that allow legal carry, but without a gun.
If you have to leave it, maybe a small, secure gunsafe in the car?
As for the traveling, I just traveled from Memphis to New Orleans through a bunch of gun-friendly county. Praise God!
Ships do indeed have scanning equipment for the luggage and you pass through metal detectors to board. Since you are traveling to international waters - they strictly enforce no guns. Also, the individual cruise lines have different rules regarding taking a knife on board so check the line before taking a sharp on board. When I go to Galveston for a cruise, I take a keltec p380 so that if somebody breaks into my vehicle and steals it, it is not a big or irreplaceable loss. I have never had a problem at the long term cruise line lots though and I don't know anyone personally that has been broken into.

If you're talking about a $500 Glock, considering what you are likely spending on the cruise, is the cost of the gun really so significant given the chances of your car being broken into are actually pretty low?
You may want to contact the cruise line to inquire about the security level of parking at the port.
Personally, I'd just leave it in the car and take my chances (even though I'll likely get flamed for it)

I know of people that have very successfully foiled thieves by putting the cable lock through the barrel of the Glock with the slide back, and locking the cable around a seat post.
Alternatively you could get a safe deposit box (make sure you have a gun rug or other small case to put the unloaded firearm into), or I know in one case of someone who was headed down to Mexico in the car and forgot he had his with him - he put the Glock in a case inside a storage locker that he rented for the month as a quick fix to avoid a long drive all the way home and back.
As mentioned - DON'T try to take it on the ship. They do scan and metal detect folks and will not be amused.
As an afterthought - check out all of the cruise line parking lots. Port of Galveston cruise line lots are fenced but are only patrolled every couple or three hours. There are also a couple of private lots that have personnel living on the fenced and lit lots. I use them. Maybe NO has something similar?

A pawn shop would be a good possibility. Pawn them for $25 each and pick them up on your return. Secured,insured and safe.
